DESCRIPTION

What It Does: Liberates hesperetin aglycone from hesperidin providing 15× enhanced tyrosinase inhibition for skin brightening — enzymatic activation providing the bioactive form with dramatically improved potency and sc penetration versus the intact glycoside.

Why It's Used: Hesperidinase converts a poorly active glycoside to a highly active aglycone — the rutinoside sugar's removal raises logp from -0.9 to +2.1 and eliminates the sugar-steric hindrance at the tyrosinase active site, providing 15× the inhibitory potency per mole.

How It Works: Hesperidinase: α-rhamnosidase cleaves rhamnosyl bond → hesperidinase-2 → glucosidase cleaves glucosyl bond → hesperetin + rutinose. hesperetin (3',5,7-trihydroxy-4'-methoxyflavone): competitive tyrosinase inhibition ic50 ~20 μm (vs hesperidin ic50 ~300 μm) from 4'-methoxy group optimal active-site positioning. logp +2.1 → improved sc penetration versus hesperidin logp -0.9.
